چکیده

A sensitive method is described for the detection of glycoproteins after polyacrylamide gel electrophoresis. Dansyl hydrazine is condensed with aldehydic functions generated by periodate oxidation of protein bound carbohydrate. The resulting hydrazones are reduced to the stable hydrazine derivatives with sodium borohydride. After destaining, the fluorescent-labeled glycoproteins are visualized with a long-range uv light source. The dansyl hydrazine staining technique was shown to detect less than 40 ng of carbohydrate bound to Bandeiraea simplicifolia lectin.

Using a Temperature Gradient against the Time in Polyacrylamide Gel Electrophoresis May Eliminate the Need for Stacking Gels

Background and Objectives: Making stacking gels for polyacrylamide gels in the laboratory by conventional methods is laborious and time consuming. Considering the role of temperature in polyacrylamide gels with respect to electrical resistance and viscosity, we assumed that decreasing the temperature would cause an increase in electrical resistance and viscosity.  Ultimately, a downward tempera...

A Novel Method to Detect β-Cyclodextrin Glucosyl Transferase (β-CGTase) Activity on Polyacrylamide Gels

b-cyclodextrin glucosyl transferase (b-CGTase) hydrolyses starch to produce b-cyclodextrin by transglycosylation (cyclization) activity. The conventional method for detection of b-CGTase activity is based on detecting starch hydrolysis by iodine staining. This method reveals all amylolytic enzymes, but can not discriminate them. In the present study, we introduce a new method for specific detec...

Simple and Specific Dual-Wavelength Excitable Dye Staining for Glycoprotein Detection in Polyacrylamide Gels and Its Application in Glycoproteomics

In this study, a commercially available fluorescent dye, Lissamine rhodamine B sulfonyl hydrazine (LRSH), was designed to specifically stain the glycoproteins in polyacrylamide gels. Through the periodate/Schiff base mechanism, the fluorescent dye readily attaches to glycoproteins and the fluorescence can be simultaneously observed under either 305 nm or 532 nm excitation therefore, the dye-sta...

Detection of proteolytic activity by fluorescent zymogram in-gel assays.

Proteases are involved in the regulation of many biological functions. This study describes a novel method for detecting protease activity by fluorescent zymogram in-gel protease assays, using SDS polyacrylamide gels copolymerized with a peptide-MCA (4-methyl-coumaryl-7-amide) substrate. This method allows simultaneous determination of protease cleavage specificity and molecular weight. Trypsin...

Visible fluorescent detection of proteins in polyacrylamide gels without staining.

2,2,2-Trichloroethanol (TCE) incorporated into polyacrylamide gels before polymerization provides fluorescent visible detection of proteins in less than 5min of total processing time. The tryptophans in proteins undergo an ultraviolet light-induced reaction with trihalocompounds to produce fluorescence in the visible range so that the protein bands can be visualized on a 300-nm transilluminator...
